Role of Nk gene Msx studied in Heart development
Msx 1 and Msx 2 (Drop ortholog from Drosophila) is a part of Nk cluster ,which display overlapping expression patterns and redundant functions in multiple tissues and organs during vertebrate development. Their role as downstream effector in BMP signalling is well characterised and to further establish MSX role in heart development Yi-Hui Chen et al, looked into Bmp-induced endothelial-mesenchymal transformation (EMT) during atrioventricular (AV) valve formation.
These Msx 1 and Msx 2 genes have redundant function in development as homozygous null mutants of individuals genes shows no phenotype as valve developed normally,but this changed dramatically in double mutants of Msx 1 and 2.Notch and Bmp signalling was reduced and hypoplastic AV cushions and malformed AV valves were observed in double null mutants of Nk Msx.Authors also suggest role of Msx genes in in regulating myocardial signals required for remodelling AV valves and maintaining an undifferentiated state of the AV myocardium.
